چکیده

Ovarian follicles are a major site of oestrogen synthesis in sheep, but almost nothing is known about the relative contribution made by follicles of different sizes to the output of oestrogen at various stages of the reproductive cycle. To determine this, ovaries were dissected from sheep, all follicles over 2 mm in diameter were explanted separately as organ cultures and the daily production of oestrogen from each follicle was measured. This study was preceded by a series of preliminary experiments undertaken to define suitable conditions in which ovarian follicles would retain both their morphological integrity and steroidogenic capacity in culture. The 224 follicles used in the preliminary series of experiments were explanted from the ovaries of seventeen sheep killed between the 2nd and 15th days of the oestrous cycle. A total of 218 follicles was used in the main experiment: 186 follicles were obtained from seventeen sheep during the oestrous cycle, seventeen follicles from two sheep at mid-pregnancy and fifteen follicles from

Background: Ovarian cryopreservation is an efficient way to preserve fertility of cancer patients before chemotherapy. This study was designed to evaluate the effect of apha lipoic acid (ALA) on reactive oxygen species (ROS) production and total antioxidant capacity (TAC) of cultured isolated pre-antral follicles derived from vitrified and non-vitrified ovarian tissue. Materials and Methods: Im...

Regeneration of atretic sheep ovarian follicles in vitro.

Large (4--6 mm diam.) and small (2--3 mm) atretic follicles were removed from sheep ovaries during the luteal phase of the cycle and maintained in organ culture without hormonal supplementation for up to 5 days. The structure, cell dynamics and steroid-producing capacity of the follicles were compared with those of non-atretic follicles of similar size. The granulosa layer of the atretic follic...

Background: Ovarian cortex xenotransplantation into immunodeficient rats is a follicles protection method in rare animals and investigation on folliclogenesis. The aim of this study was to evaluate the effects of gonadotropin and erythropoietin (EPO) treatment on follicular development in the transplanted ovaries. Materials and Methods: Prepared sheep ovarian cortical strips were vitrified by c...

Biochemical and hormonal characterization of follicles from follicular and luteal phase ovaries of goat and sheep.

Follicles from goat and sheep ovaries were characterized for their biochemical and hormonal parameters to investigate the effect of developmental stage of follicles on ovarian steroidogenesis. The follicles were isolated mechanically from follicular and luteal phase ovaries and divided in 6 morphologically different groups (small, medium and large follicular and small, medium and large luteal)....
